No instructions needed to laugh
I came to this podcast by way of the Knights' live show; and knowing no more about D&D than what I saw on "Community," had a blast.
I was afraid that the podcast wouldn't live up to the magic in the room...but I was wrong. Knights of the Arcade is the best value show in Pittsburgh and across the Interwebs.
Do you get hung up on minutiae, and will it bother your OCD if they venture off of the game rules to tell some extra fart jokes? Then maybe skip it and go back to the dungeon.
If you enjoy ridiculous characters, just enough of a narrative structure to keep the gang from getting stuck in the improv mire, and the occasional dice roll completely screwing things up (and the occasional song), you should really give this a go. I can't say enough about them.
Also, I enjoyed their first detour (podcast-wise, at least) into board games. That's just a personal preference, but I feel like there's enough overlap between the groups to keep us engaged while we wait for the next full episode/quest (and doing Betrayal at Baldur's Gate is a perfect crossover).

Do you like laughing?
The show and the podcast are great! I try to make the show whenever possible, and listen to the podcast when I can't. Heck, I listen to the podcast when I do make it. I don't play D&D but this is still the best show at the Arcade!
I am also REALLY excited they are branching into board games. I love board games, I love the knights! It's a winning combo.
You don't need to love or understand either of those things to love this podcast though. Do you like laughing? Then this is for you!

Knights of the Arcade is hliarious! I can't believe I'm just getting on board with this great podcast. Super accessible for everyone, not just D & D players. Burt Conroy is awesome... only second in awesomeness to the late, great Stone Cold.

Fills a perfect niche
Really offers something I hadn't seen in another podcast. Unlike the countless "some friends decided to record their D&D sessions" podcasts, this one is performed in front of an audience with improv comedians whose goal it is to entertain their audience, not delve into rules minutia.

Great time at the Arcade! Highly recommend for even the most casual of gamer. All laughs, all cannon, and surprisingly talented bards. Can't beat it.

An awesome evening
This show is fantastic. I've listened to a lot of table-top podcasts and this is something special. Each show being a self contained quest is a great idea for the format, and having the audience come up with that quest is a nice touch.
I went down to see Spencer (who did an amazing job as the guest DM in episode 3) having no idea what to expect. I was blown away. The party memebers are all wonderfully funny and quick-witted. They've got a projector set up so you can see the rolls as they happen. They've got a bard who sits behind a keyboard, improvising all his musical rituals on the spot. They throw beer and chips into the crowd everytime they crit. I mean, what more could you ask for?
If you're anywhere near Pittsburgh you've got to go see this show. If not, the podcast will have to do, you'll just have to provide your own chips and beer.

What happens when you gather a group of comedians, special guests and a nerdy audience and thrust them into a fantasy world of mayhem and magic? Probably something awful, so instead listen to this hilarious podcast which simulates those elements to hysterical effect.
